My daughter is now studying in P4 local English primary school, however, I am thinking about changing her to HKIS at P6 which is G6 at HKIS because we will be moving to the USA before her high school. Should I apply now and put her on waiting list or should I apply when the time comes? Would she get a better change being admitted at G6 or earlier since G6 is the first year of their middle school? Can anyone experienced this or have information about my questions? Thank you.

If you want your daughter to get into HKIS, you'd better apply now as HKIS always has a long waiting list unless you got some sorta priority in the admission process.

I think there is no such saying of "having a better chance to apply before the first year of middle school" but what is the best for your daughter. Frankly, I don't know much about middle school admission but when we look into high schools, you would notice that many students would consider apply to US high school at G9 instead of applying G7 or 8 to "get a better chance of getting into the high school."

My view is, apply as soon as possible. It is not up to you to decide when you want to go but it totally depends on the availability of places.
Then in the mean time, do your due diligence and search for high schools in the US. Good private schools are extremely hard to get a place.
You might also want to consider ICS as a backup, they also adopt the US curriculum and I heard they are a decent school.
